Identify HLA-binding peptides from a whole protein
The Epibase® MAPPs (MHC-associated peptide proteomics) assay identifies HLA-binding peptides naturally processed and presented from a whole biotherapeutic protein. The regions identified can be targeted for removal of potential T cell epitopes or ‘deimmunization’.
MAPPs Assay
- Dendritic cells (DCs) are generated from healthy donors and loaded with test biotherapeutic protein
- DCs take-up, process and present peptides from the protein via HLA Class II molecules
- DC lysis and immunoprecipitation of HLA-DR molecules
- Elution of peptides from HLA-DR molecules
- Peptides identified by mass spectrometry and mapped to biotherapeutic protein sequence
- Internal proteins included as controls
- Immunogenicity risk of identified peptides predicted
